Nació el 14 de julio de 1926 en West Irvine, Kentucky, Estados Unidos.

Hijo de Sheridan Harry Stanton, agricultor de tabaco y barbero, y de Ersel Moberly, peluquera. Sus padres se divorciaron cuando él estaba en la escuela secundaria; los dos se volverían a casar. Tiene dos hermanos más jóvenes, Archie y Ralph, y un hermanastro, Stan.

Asistió a Lafayette High School y a la Universidad de Kentucky en Lexington, donde estudió periodismo y actuó en el Guignol Theatre bajo la dirección del icono del cine del Reino Unido Wallace Briggs. Después estudió en el Pasadena Playhouse, en Pasadena, California, donde tuvo a compañeros de clase como Tyler MacDuff y Dana Andrews, convirtiéndose en un actor muy prolífico apareciendo en películas de cualquier género.

A lo largo de 60 años, trabajó para directores como Francis Ford Coppola en El Padrino: Parte II; Sam Peckinpah en Pat Garrett y Billy The Kid; Martin Scorsese en La última tentación de Cristo o Ridley Scott en Alien. Secundario característico especializado en papeles de inadaptado, vagabundo o personajes al margen, Wim Wenders le dio su gran oportunidad como protagonista en Paris, Texas.

Desde 2006 encarnó a Roman Grant en la serie Big Love de HBO. Participó en más de 70 películas y en muchas series de televisión, entre ellas Twin Peaks, de David Lynch, que también lo dirigió en Wild at heart, Inland Empire y The Straight Story,

Participó en una banda llamada "The Harry Dean Stanton Band" con su guitarra. Participó en el video musical Dreaming of You de Bob Dylan.

Harry Dean Stanton, falleció el viernes 15 de septiembre de 2017 en el hospital Cedars Sinai de Los Ángeles de causas naturales.
